Design: Whole insect was digested in custom lysis solution with Proteinaze K and DNA was extraxted using magnetic beads. Libraries were prepared using a two-step PCR approach. For the first PCR combination of template-specific primers with adapter stubs were used: for the V4 region of 16S rRNA gene and for a partial region of the insect mitochondrial COI gene. After the bead-purification, products were completed with unique Illumina adapters in second PCR. In both PCR reaction Qiagen Multiplex PCR Master Mix polymerase was used.
